Measured on two holes per round, usually two relatively flat holes with wider fairways, and that run in opposite directions to account for any wind effects.
I don't see the stat that shows Partrick number one for the season, and that number you cited is too high. That must be two drives in a particular round, or something--or a misprint. Partick is pretty steady about 305-310, and i am all but certain that Dustin Johnson leads (around 315 ypd) based on a reasonable minimum number of events entered.
